📢[New Publication!] 💉Did you know that adeno-associated virus (AAVs) are emerging as promising tools for gene therapy applications? One of the limiting factors in #AAV production process is the poor efficiency of the #purification step. AAVs are usually purified using affinity and ion exchange #chromatography with packed beds of resin absorbents. However, this often leads to diffusion and pressure drop limitations, affecting overall process productivity. 📄In this new publication, iBET scientists in partnership with Cytiva, investigated the use of affinity cellulose-based #nanofiber absorbents for AAV serotype 5 capture. ➡️This approach enabled a 10-fold reduction in residence time in comparison with packed-bed chromatography, with similar AAV recovery and impurity removal. Scientists have taken a significant step forward in developing a robust measles virus pseudotyped lentiviral vector (MV-LV) protocol! Their research showed enhanced MV-LV mediated efficient transduction of primary human B cells, providing assays to quantify MV-LV titers, B cell transduction efficiency and VCN. Flow cytometry was also used to characterize primary B cell phenotypic characteristics. This was a major breakthrough in understanding the potential of MV-LV vectors for gene delivery. BMKMANU S1000 Spatial Transcriptomics Project Experience - Olfactory Bulb in Mouse Brain Using single-cell level spatial transcriptome technology, BMKMANU S1000, the olfactory bulb sample in the mouse brain was detected, obtaining a total of transcripts and spatial positional information of 17,564 cells. The BMKMANU cell segmentation technology was used to accurately identify the different types of cells in the olfactory bulb sample. This helped in reconstructing the spatial distribution of various layers such as the nerve layer, glomerular layer, external plexiform layer, internal plexiform layer, mitral valve layer, granule layer, and subependymal zone in the main olfactory bulb.
