The Broad Institute brings together a diverse group of individuals from across its partner institutions — undergraduate and graduate students, postdoctoral fellows, professional scientists, administrative professionals, and academic faculty. The culture and environment at the Broad is designed to encourage creativity and to engage all participants, regardless of role or seniority, in the mission of the Institute. Within this setting, researchers are empowered — both intellectually and technically — to confront even the most difficult biomedical challenges. The Institute’s organization is unique among biomedical research institutions. It encompasses three types of organizational units: core member laboratories, programs and platforms. Scientists within these units work closely together — and with other collaborators around the world — to tackle critical problems in human biology and disease.

Thank you to everyone who joined us for our recent Broad Discovery Series event, “Moving from "hit-or-miss" toward a brighter future for psychiatric diagnosis and care.” The panel discussion focused on the topic of mental health and the challenges faced in psychiatric diagnosis and care. Key themes that emerged from the conversation were the importance of support and community, the need for more objective and early diagnoses, the role of genetics in understanding mental illness, and the potential for personalized and holistic treatment approaches. Our panelists emphasized the need for inclusivity and diversity in research and the importance of hope in driving progress. Also highlighted was the significance of lived experiences in shaping research direction and the value of advocacy groups like NAMI in supporting individuals and influencing research. Overall, the discussion emphasized the ongoing efforts to improve mental health care and the potential for advancements in the field. Epstein-Barr virus (EBV) infects 95 percent of adults worldwide and can trigger multiple sclerosis and cause cancer. To maintain infection throughout a person’s life, the virus has a two-phase life cycle consisting of periods of latency (no replication) and lytic reactivation (active replication and release of new viral copies). Scientists don’t yet know which host factors govern EBV latency. In Nature Communications, Laura Murray-Nerger, Ph.D., Ben Gewurz, and colleagues describe using human genome-wide CRISPR screens to find that a protein in the nucleus called SFPQ controls the expression of histone H1, which is critical for the latent period. The heart's right atrium is a crucial structure that hasn’t been well studied in the context of chronic heart diseases. Suvi Linna-Kuosmanen, Eloi Schmauch, Manolis Kellis, and colleagues report in Cell Reports Medicine a comprehensive single-cell and spatial transcriptomic atlas of the human right atrium in valvular and ischemic heart disease progressing to heart failure. They found substantial differences in disease-associated gene expression in all cell types that suggested inflammatory microvascular dysfunction and changes in tissue composition during disease progression. Diabetic kidney disease (DKD) is a common complication of diabetes and obesity, and lacks effective treatments. Ayshwarya Subramanian, Katherine Vernon, Yiming Zhou, Anna Greka, MD, PhD, and colleagues analyzed single-cell transcriptomic profiles from DKD patients and two DKD mouse models at multiple time points during disease progression. In Cell Reports, they describe an expanding population of macrophages expressing high levels of TREM2 in mice fed a high-fat diet and show that human diabetic and obese kidney tissue harbored higher levels of TREM2 macrophages. Deleting Trem2 in mice promoted kidney injury after exposure to a high-fat diet. Big congratulations to Boston Children’s own Stuart Orkin, MD, for receiving The Shaw Prize in Life Science and Medicine. The 2024 prize is shared with Swee Lay Thein of the Sickle Cell Branch, National Heart, Lung, and Blood Institute. The prize recognizes their work on understanding the fetal-to-adult hemoglobin switch — and suppressing it by targeting the BCL11A gene in people with sickle cell disease and beta thalassemia. Liver cells robustly adapt to nutrient fluctuations, but the subcellular remodeling involved isn’t fully understood. Gunes Parlakgul, M.D., Gökhan Hotamışlıgil, Ana Paula Arruda, and colleagues analyzed the spatial organization of mouse liver cells' organelles as nutrients fluctuate. During fasting, the endoplasmic reticulum wrapped mitochondria in large flat sheets within cells from some of the liver’s zones but not others, in a process dependent on RRBP1. In cells from obese mice, fasting failed to produce the same subcellular changes.
